Spin-column kits purify DNA on a silica membrane. In the presence of high salt, DNA binds to the silica while proteins and other contaminants pass through.
The workflow has four steps: lyse the sample to release DNA, bind it to the column, wash away contaminants with ethanol-based wash buffers, and elute pure DNA in a small volume of elution buffer.
A few details make a big difference. Add ethanol to the wash buffers before first use. Pre-heat the elution buffer when the protocol asks for it. Spin at the recommended speed so the membrane dries fully before elution.
Helix kits cover blood, cultured cells, tissue and plant samples, as well as plasmid preparation and gel or PCR clean-up, each with a protocol and troubleshooting guide.
