RNA is easily degraded by RNases, enzymes that are found on skin, dust and lab surfaces. Even trace amounts can reduce yield and integrity and give inconsistent results in RT-PCR or Northern blotting.
Start with the basics: wear gloves and change them often, use RNase-free tips and tubes, and keep a separate, clean area for RNA work.
Decontaminate surfaces and pipettors before you start. A solution such as Helix Nuclease Terminator is sprayed on the surface, left for 30 seconds to 5 minutes, and wiped off with lint-free tissue wetted with nuclease-free water.
Finally, add an RNase inhibitor to sensitive reactions such as cDNA synthesis, and store purified RNA at -80 °C.
